The objective of this book is to present protocols, methodologies and standard operating procedures (SOPs) for measuring GHGs from different agroecosystems using isotopic and related techniques, which can also be used to validate climate-smart agricultural practices to mitigate GHGs. The material presented in this book should be useful for both, beginners in the field, to obtain an overview of the current methodology, and experienced researchers who need a hands-on description of current methodologies. We hope that the methods described in this book are easy-to-understand and applicable to a range of users with different expertise and backgrounds. This book is an outcome of the ollaboration between the Soil and Water Management and Crop Nutrition Section, Joint FAO/IAEA Division of Nuclear Techniques in Food and Agriculture, Department of Nuclear Sciences and Applications, International Atomic Energy Agency (IAEA), Vienna, Austria and the German Science Foundation research unit DASIM (Denitrification in Agricultural Soils: Integrated control and Modelling at various scales) among other institutes. DASIM is a consortium of scientists in Germany with associated partners worldwide which studies, in detail, the processes of GHG emissions using nuclear and stable isotopic techniques of 15N, 18O and 13C to understand and quantify gaseous N fluxes. This information has been compiled from the latest published literature and from authors? own publications specific to the subject matter (especially with expertise coming from the DASIM network). O milho é um dos cereais mais cultivados no Brasil. Além de ser um componente das rações animais, também constitui um item importante da dieta brasileira. O maior problema desta cultura é a presença de toxinas fúngicas, que reduzem a sua qualidade. Para sanar esta situação, deve haver um controle de qualidade adequado. Tal controle demanda por análises do tipo multianalito para quantificar as micotoxinas presentes no milho. Este trabalho apresenta um método para análise simultânea de treze micotoxinas: aflatoxinas B1, B2, G1 e G2; fumonisinas B1 e B2, ocratoxina A; nivalenol; desoxinivalenol; zearalenona; toxina T2; toxina HT2 e esterigmatocistina no milho por cromatografia líquida de ultraeficiência acoplada à espectrometria de massas sequencial (CLUE-EM/EM). O método foi validado e se mostrou seletivo, exato e preciso dentro da faixa de trabalho avaliada, com limites de detecção e quantificação inferiores ao limite máximo tolerado (LMT) de cada micotoxina no milho. |
