The nuclear gene Msh1 (MutS homologue 1), recently cloned in Arabidopsis (Abdelnoor et al., 2003, PNAS 100:5968-5973), is involved in regulation of the copy number of subgenomic mitochondrial DNA molecules created by ectopic recombination in a process known as substoichiometric shifting. The Arabidopsis genome contains several other MutS homologues, but MSH1 protein is the only one that is targeted to both mitochondria and chloroplast and the predicted computer-based structure of this protein is very similar to the E. coli MUTS. We have isolated and sequenced the full-length homologous Msh1 genes in soybean, rice, tomato, and common bean to better understand their structure and function in plants. The tomato and soybean genes were obtained based on EST sequences identified by similarity to the Arabidopsis Msh1 gene and by RT-PCR of total RNA from leaves. The rice Msh1 was identified by searching the rice BAC sequences database and the coding sequence was confirmed by RT-PCR. The bean Msh1 was isolated from a BAC library, using a sequence of the soybean Msh1 gene as probe. Similar to Arabidopsis Msh1, all of them are comprised of 22 exons with very well conserved size and splicing position. Phylogenetic analysis using ClustalW showed that these genes have an overall identity of about 60% at the protein level. Based on computer prediction analysis they are predicted to be targeted to mitochondria. Additionally, several other ESTs have been identified in other plant species (barley, wheat, potato, tobacco, sorghum, maize and medicago), and show high homology to the sequenced Msh1 genes. The alignment of all of these MSH1 proteins has allowed us to identify specific conserved domains that might be very important for their proper function in plant mitochondria. O objetivo deste trabalho foi avaliar a viabilidade do emprego simultâneo de genes mutantes de amadurecimento e de coloração, nos locos norA, rin, ogc e hp, em híbridos de tomateiro, com diferentes backgrounds e combinações genotípicas. O experimento foi conduzido em estufas, em delineamento de blocos ao acaso, com 20 genótipos e três repetições. Houve tendência dos mutantes de amadurecimento (nor+/norA e rin+/rin) para reduzir a produção precoce de frutos, mas não a produção total. Genótipos portadores de um ou mais alelos norA, rin, hp e/ou ogc apresentaram maior conservação de frutos na pós-colheita que genótipos normais. Genótipos rin+/rin apresentaram as piores colorações internas de frutos. Houve um efeito positivo de ogc+/ogc, ogc/ogc e/ou hp+/hp na coloração interna de genótipos normais, rin+/rin e nor+/norA, mas o efeito em genótipos rin+/rin não foi de magnitude suficiente para tornar a coloração interna final semelhante à de genótipos normais. As constituições genotípicas ogc+/ogc e ogc/ogc apresentaram efeitos semelhantes na melhoria da coloração interna dos frutos e da coloração da mucilagem placentária. As constituições genotípicas nor+/norA ogc+/ogc ou nor+/norA ogc+/ogc hp+/hp foram consideradas as mais promissoras, tanto para melhorar a conservação pós-colheita, quanto para manter ou melhorar a coloração interna dos frutos do tomateiro. |
