Tospoviruses are the only plant-infecting members of theBunya-viridaefamily of ambisense ssRNA viruses. Tomato spotted wilttospovirus (TSWV), the type-member, also causes mild infection onits main insect vector,Frankliniella occidentalis. Herein, we iden-tified anF. occidentalisputative transcription factor (FoTF) thatbinds to the TSWV RNA-dependent RNA polymerase and to viralRNA. Usingin vitroRNA synthesis assays, we show that addition ofpurified FoTF improves viral replication, but not transcription.Expression of FoTF deletion mutants, unable to bind the RNA-dependent RNA polymerase or viral RNA, blocks TSWV replicationinF. occidentaliscells. Finally, expression of FoTF wild-type turnshuman cell lines permissive to TSWV replication. These data indi-cate that FoTF is a host factor required for TSWV replicationin vitroandin vivo, provide an experimental system that could be used tocompare molecular defense mechanisms in plant, insect, and hu-man cells against the same pathogen (TSWV), and could lead to abetter understanding of evolutionary processes of ambisense RNAviruses. |
