Abstract: The effects of freezing and thawing upon ovine spermatozoal acid phosphatase, lactate dehydrogenase, and deoxyribonucleic acid were determined. Two bands of seminal acid phosphatase (P-AP1 and P-AP2) and two bands of spermatozoal acid phosphatase (S-AP1 and S-AP2) were detected by acrylamide gel electrophoresis. Seven bands of both seminal and spermatozoal lactate dehydrogenase activity were also observed. Freezing and thawing produced a significant increase in the activity of P-AP2 band of seminal plasma and a concomitant significant decrease in the activity of S-AP2 band in spermatozoa (P < 0.05). This suggests that some S-AP2 isozyme leaks out of the sperm into the diluent. The three lactate dehydrogenase isozymes measured did not show alterations in activity following freezing, thawing, or incubation. The deoxyribonucleic acid content of ram spermatozoa remained unaffected by freezing, thawing, or incubation. |
