Two bands of seminal acid phosphatase (P-AP1 and P-AP2) and two bands of spermatozoal acid phosphatase (S-AP1 and S-AP2) were detected by acrylamide gel electrophoresis. Seven bands of both seminal and spermatozoal lactate dehydrogenase activity were also observed. Freezing and thawing produced a significant increase in the activity of P-AP2 band of seminal plasma and a concomitant significant decrease in the activity of S-AP2 band in spermatozoa (P < 0.05). This suggests that some S-AP2 isozyme leaks out of the sperm into the diluent. The three lactate dehydrogenase isozymes measured did not show alterations in activity following freezing, thawing, or incubation.
