A deep freezing technique of optimal semen obtained with an artificial vagina, from 4 male Nubian goats (3-5 years old) and diluted 1:4-1:8 with Laiciphos 271, assuring a 200-300 x 10 motile spermatozoa concentration perfrozen dose and 100-200 x 10 after thawing, is described. The addition of 4 vol. % glycerol was undertaken inin three steps. The first one 2 % in the initial diluent at 30-32o C (50% of the final volume) and the other two at 6% in the final dilution at 20o. y 5o C respectively (25% of the final volume each time). The cooling at 5o C was made in 30-45 minutes, with an equilibration time of 60 minutes. Samples of seen were frozen horizontally in medium size straws (0.5 ml) for 5 minutes, 4 cm above the surface of the liquid N2, and thawed in 10 seconds in water at 75o C. Samples below 60% of progressive motility were discarded. In 35 native goats, intracervical insemination was perfomance twice during estrual period 68.6 % conceived during the first period and 14.4 % in a second period, with a total of 82.8% and 1.4 kidding per goat. To overcome rapid lose of fertility of liquid semen, frozen semen is reccomended for crossbreeding in caprine herds. |
