Here, bioinformatic approach based on protein modeling was applied to two ORFs related to ketogluconates production, gluconate 5-dehydrogenase and 2-ketoglucone reductase. The ORFs were analyzed and identification of regions that play an important role in substrate binding/recognition presents an opportunity for stability prediction and design of mutagenesis strategy. Gluconate-5-dehydrogenase was modelled using as a 3D model the crystal structure of Gluconate 5-dehydrogenase from Thermotoga maritima (PDB ID: vl8) whereas crystal structure of a NDA-dependent p-glycerate dehydrogenase (PDB ID:1gdh) was used to model 2-ketogluconate reductase. The sequence identities for these proteins are 36% and 34%, respectively. Based on this in silico model and with the prediction performed with CUPSAT [4], it was proposed to construct the mutants His289Asn and Glu271Ser of 2-ketoglucone reductase. |
