A complementary DNA library was constructed to the genomic RNA of an isolate of papaya ringspot virus (PRSV), PRSV YK (YK), that represents the most prevalent mosaic strain of Taiwan. A 2.96-kb clone, pYKB9, corresponding to the 3'-region of YK RNA was selected by immunoscreening with the antiserum to the coat protein (CP). The clone had 2,960 nucleotide residuesand represented part of the nuclear inclusion a (NLa) gene, the complete nuclear inclusion b (NLb) gene, the entire CP gene, and the 3'-noncoding region of YK RNA. The nucleotide sequence was compared to those of the 3' regions of the published Hawaii severe strain, PRSV HA (HA), the Hawaii mild strain PRSV HA 5-1 (HA 5-1), the monpapaya-infecting Florida strain, PRSV W(fl) (W(fl), and the monpapaya-infecting Australia strain PRSV W(aust) (W(aust)). The NIb region had 83.1% nucleotide identity and 95.7% amino acid identity compared to thatof HA, with a total of 262 nucleotide differences resulting in only 22 amino acid changes. A similar trend also was found when the NIb region of YK was compared to the partially elucidated NIb regions of HA 5-1and PRSV W(fl), with 84.7 and 85.2% nucleotide identities (147 and 151 nucleotide differences), respectively: both resulting in 97.6% amino acid identity (only eight amino acid changes). Most of the nucleotide differences between the Asian and American strains occurred in wobble bases and, thus, conserved the amino acid residues, suggesting a common selection pressure to keep the same protein conformation. A indústria de panificação tem sofrido uma revolução nos últimos anos, buscando novas mesclas de farinhas de forma a oferecer novas variedades de pães com diferentes características sensoriais. A farinha de pinhão pura pode ser utilizada de forma diversa na indústria de panificação, dada a sua praticidade, estabilidade, sabor suave e textura fina, o que possibilita a geração de novos produtos. Entretanto, as farinhas mistas, quando adicionadas à massa do pão, podem ocasionar alterações na consistência, textura, volume e comportamento reológico, comprometendo em alguns casos a aceitação do produto final devido às modificações de sabor, redução do volume, aumento na dureza e escurecimento da massa. O objetivo desse estudo foi avaliar as características reológicas da mescla de farinha de trigo com farinha de pinhão em diferentes proporções (0%, 5%, 10%, 15%, 20% e 25%). Houve diferenças significativas nos parâmetros de absorção, tempo de desenvolvimento da massa, índice de tolerância à mistura, estabilidade, força e número de queda. A mistura de farinha de trigo e de pinhão aumentou o teor de fibra bruta e reduziu a qualidade tecnológica como estabilidade, extensibilidade, força, tempo de desenvolvimento da massa e aumento da tenacidade pelo índice de tolerância à mistura. Essas alterações foram verificadas quando a proporção de farinha de pinhão na mescla de farinhas foi superior a 10%. Portanto, os melhores resultados são obtidos com mesclas de até 10% de farinha de pinhão. |
