Abstract: Fusarium graminearum is the main cause of Fusarium head blight, a fungal disease that reduces yield and afects the quality of wheat and other small grains. Genetic resistance is the ideal method for disease control; however, there are not yet cultivars with sufcient resistance levels to withstand an epidemic. Genetic engineering strategies such as gene silencing, host-induced gene silencing (HIGS), overexpression, and genome editing are promising, but many target genes need to be analyzed to fnd a suitable one. Arabidopsis thaliana is a model plant that is also susceptible to F. graminearum. Although this interaction was reported nearly two decades ago, consistent infection and symptoms are not always obtained. The availability of an efcient inoculation method is essential for studying plant-pathogen interaction. This work aimed at testing protocols for inoculating F. graminearum in detached leaves of A. thaliana which varied in inoculation site (leaf sides), wound (abrasion or hole injury), and inoculum type (mycelium agar disk or spore). We found that a mycelium agar disk placed on an abrasion injury at the adaxial leaf side was the most efcient (highest lesion size, incidence, and easiness to visualize the symptoms caused by the pathogen) method to inoculate F. graminearum in detached leaves of Arabidopsis. |

RESUMO: Este trabalho foi conduzido no viveiro da Embrapa Cerrados, em Planaltina (DF), com a reealização da enxertaria durante a estação chuvosa de verão. Teve como objetivo avaliar a viabilidade da enxertaria de mudas de pequizeiro (Caryocar brasiliense Camb.) pelos métodos de garfagem lateral ou à inglesa simples, borbulhia de placas sem lenho e com janela aberta e borbulhia de escudo com lenho. Os porta-enxertos foram produzidos em sacos plásticos de 20 x 30 cm e possuíam diâmetro do caule acima de 0,7 cm a 10 cm do solo. Para o fornecimento dos enxertos, foram selecionadas quatro plantas adultas, em bom estado vegetativo, com folhas maduras nas últimas brotações e sem frutos. O delineamento experimental foi o de blocos ao acaso, com três tratamentos (três métodos de enxertia) e oito repetições de dez enxertos (duas repetições por planta-matriz, correspondendo a oito blocos). A abertura (retirada da fita) dos enxertos foi feita depois de 30 e 90 dias da borbulhia e da garfagem, respectivamente, porém, a avaliação do pegamento desses enxertos foi realizada aos três meses, computando-se apenas aqueles vivos e os brotados. Os resultados evidenciaram a viabilidade da clonagem de pequizeiros adultos pelos métodos de garfagem lateral e borbulhia de placa sem lenho e com janela aberta os quais proporcionaram em torno de 60% e 90% dos enxertos pegos, respectivamente. A enxertia por borbulhia de escudo com lenho é inviável, não se constatando enxertos vivos. ABSTRACT: This work was carried out int he nursery at the Embrapa Cerrados, in Planaltina, Federal District, Brazil. The objective was evaluate the vegetative propagation of pequi (na edible friut specie from the Brazilian Savannah) by the grafting and budding methods (spliced side graft, pacth budding and chip budding). Seeding rootstocks were grown in polybags of 20 cm x 30cm and they had stem diameter of at least 0,7 cm at 10 cm above the ground. Both the grafts and the budsticks were taken from four adult fruitless plants with mature leaves in the last apical leaf flow. The experimental desing was in randomized complete blocks, with eight replicates of ten plants per plot (two replicates per source plant). The wrapping tape was removed about 30 or 90 days after budding or grafting, respectively, and the final evaluation of bud and graft success was made in the latest epoch, taking in count only the live and sprouted scions. Both the spliced side graft and the pacth budding methods showed to be promising for the vegetative propagation of pequi, presenting graft and bud success of about 60% and 90%, respectively. No bud success was observed for the chip budding method.
