Lung sarnples from pneumonic lesions in cattle and goats, natura1ly or experimentally infected with strains of the Mycoplasma mycoides cluster, were fixed in formalin and embedded in paraffin. An immunohistochemical technique using monoclonal or polyclonal antibodies was performed on tissue sections in order to detect Mycoplasma antigens. Four monoclonal antibodies (MAbs), one (2A3) raised against M. mycoides ssp. mycoides small colony (SC) and large colony (LC), twO (ID3 and SES) against M. mycoides ssp. capri, and one (5A10) against M. bovis, were used. A range of polyclonal antibodies, raised to the individual subspecies of the M. mycoides cluster, and one to Pas/eure/la haemo!ytica, was also used. The MAb 2A3 showed positive immunostaining in lung sections from cattle and goats natura1ly and experimentally infected with M. mycoides ssp. mycoides SC and LC, but not with pneumonic lesions of cattle and goats due to other members of the M. mycoidts cluster, M. bovis or Pasteure/la spp. The MAb ID3 showed immunostaining in lung sections from goats naturally and experimentally infected with M. mycoides ssp. capri, but again not with pneumonic lesions caused by other members of the M. mycoidts cluster, M. bovis or Pasteurella spp. The MAb SES immunoreacted in sections from pneumonic lesions from ail animais infected with one of the three M.mycoides cluster subspecies used in the study, but not with M bovis or Pasteurella infected tissue. lmmunoreaction was mainly found in the cell debris around necrotic areas, as well as in macrophages, neutrophils and epithelial cells. The localization of antigens of the M. mycoides cluster using polyclonal antisera followed basically the same pattern as mat obtained with the monoclonals. However, a wide cross reactivity was found between different antisera and relatively high background immunostaining was also seen, especially in necrotic areas. The results suggest that immunohistochemical methods using monoclonal antibodies are useful tools for the diagnosis and study of the pathogenesis of pneumonia caused by the Mycoplasmas of the M. mycoides cluster. Aims: We report the first occurrence of an epizootic of the ascomycete fungus, Isaria tenuipes (teleomorph Cordyceps takaomontana), on the ocola skipper Panoquina ocola (Lepidoptera: Hesperiidae), an insect pest affecting rice in Brazil. Methods and Results: Field surveys in flooded rice fields in the state of Goias in Brazil were conducted, and a fungal pathogen of a caterpillar pest (P. ocola) was serendipitously found. This fungus inflicted high infection levels (average 86-2%) mainly to the pupal stage during warm, humid growing conditions. Typically, mycosed pupal cadavers produced milky-white to pale yellow synnemata bearing an average of 1-1 + 0-2 x 109 conidia per cadaver. Based on phylogenetic analysis using beta-tubulin gene sequences, we confirmed that all 12 isolates obtained from field-mummified pupae were identifiable as I. tenuipes. Conclusions: Our data expand our knowledge on the host and geographical ranges of this mycopathogen and underscore its epizootic potential to affect a lepidopterous insect pest on rice in Brazil. This finding may facilitate the exploitation of this fungus as a mycoinsecticide. Significance and Impact of the Study: Isaria tenuipes may be used as an effective and environmentally friendly bioinsecticide against agricultural caterpillar pests due to its epizootic potential, as well as explored for medicinal purposes by pharmaceutical industry. |
