Clay minerals are suitable matrices to anchor organic molecules such as antimicrobial peptides (AMPs) so that their bioactivity is maintained, enabling the formation of new materials with potential for new applications in biotechnology. The objective of the present study was to develop a nanostructured flm where the properties of palygorskite (Plg) were combined at the molecular level with Dermaseptin 01 (DRS 01), in which the clay mineral also served as a substrate for the immobilization of this peptide. The flms were prepared using the Layer-byLayer (LbL) self-assembly technique. Crude palygorskite without purifcation (Plg IN) was subjected to physical and chemical procedures to increase its adsorptive properties. The structure, chemical composition, and morphology of Plg were investigated by X-ray difraction (XRD), Fourier-transform infrared spectroscopy (FTIR), X-ray fuorescence spectrometry (XRF), scanning electron microscopy (SEM), and transmission electron microscopy (TEM). LbL flms were adsorbed onto ITO (Indium Tin Oxide) and characterized electrochemically by cyclic voltammetry (CV), UV-Visible spectroscopy, and atomic force microscopy (AFM). For the ITO/DRS 01 and ITO/Plg/DRS 01 flms, an oxidation process at +0.77 V was observed, confrming that the DRS 01 maintained its electroactive behavior and intrinsic properties. The results also showed that Plg served as excellent support for the immobilization of DRS 01, increasing its concentration and availability in the flm form. This work reported immobilizing the DRS 01 peptide with Plg for the frst time in an ultrathin flm with bioactive properties. Thus, the flm developed can be explored for applications such as biosensor devices and antimicrobial coating materials as well as other biotechnological applications. O objetivo deste estudo foi determinar a extensão em que a altura da planta e da cobertura do solo poderia ser usada como indicador da produção de pastagem em capim-elefante, sob pastejo. Amostras da produção de pastagem, altura da planta e cobertura do solo foram realizadas em pastagem de capim-elefante (Pennisetum purpureum Schum.) durante os anos de 1992, 1993 e 1994. Foram usadas 30, 72 e 72 amostras para cada ano de avaliação, respectivamente. Os dados da produção de matéria seca foram submetidos à análise de regressão, em função da altura da planta e da cobertura do solo, usando vários modelos. O modelo linear foi o que apresentou o melhor ajuste dos dados, tanto para cobertura do solo quanto para o índice altura da planta vs cobertura do solo. As melhores estimativas da produção de matéria seca foram obtidas quando se combinaram medidas de altura da planta com as de cobertura do solo, analisadas por meio de regressão múltipla. Estas regressões revelaram valores de R2 mais elevados, indicando que a altura da planta e a cobertura do solo podem ser consideradas características complementares, como estimadores da produção de matéria seca em pastagem de capim-elefante. |
