Abstract: Biological nitrogen fixation is a key process for agriculture production and environmental sustainability, but there are comparatively few studies of symbionts of tropical pasture legumes, as well as few described Bradyrhizobium species, although it is the predominant 48 genus in the tropics. A detailed polyphasic study was conducted with two Bradyrhizobium strains used in commercial inoculants for tropical pastures in Brazil, CNPSo 1112T isolated from perennial soybean (Neonotonia wightii), and CNPSo 2833T from desmodium (Desmodium heterocarpon). Based on 16S-rRNA phylogeny, both strains were grouped in the B. elkanii superclade, but were not clearly clustered with any known species. MLSA of three (glnII, gyrB and recA) and five (+ atpD and dnaK) housekeeping genes confirmed that the strains are positioned in two distinct clades. Comparison with ITS sequences of described Bradyrhizobium species showed similarity lower than 93.1%, and differences were confirmed by BOX-PCR analysis. Nucleotide identity of three housekeeping genes with described species ranged from 88.1% to 96.2%. ANI of genome sequences showed values below the threshold of Bradyrhizobium species (? 90.6%) and also between the two strains (91.2%). The analysis of nifH and nodC genes positioned the two strains in a distinct clade from other Bradyrhizobium species. Morpho-physiological, genotypic and genomic data supported the description of two novel clades in the genus Bradyrhizobium, B. tropiciagri sp. nov. (type strain CNPSo 1112T =SMS 303T =BR 1009T =SEMIA 6148T =LMG 28867T) and B. embrapense sp. nov. (type strain CNPSo 2833T =CIAT 2372T =BR 2212T =SEMIA 6208T =U674T). O objetivo do trabalho foi avaliar o efeito da adição de um concentrado zeolítico enriquecido com N, P e K ao substrato de cultivo sobre o crescimento, produção de matéria seca, área foliar, teores e extração de N, P e K e os teores de clorofila do limoeiro "Cravo". O porta-enxerto foi cultivado por 93 dias em tubetes de 150 cm3 com substrato orgânico compostado de casca de coco e carvão vegetal (3:1) ao qual se adicionou o concentrado zeolítico. Este foi obtido com a concentração da zeólita natural (Z) e enriquecimento desta com KNO3 (ZNK), e também com a acidificação com H3PO4 e mistura com apatita (ZP). Utilizou-se uma mistura de 30%ZNK + 70%ZP nas doses de: 0; 2,5; 5; 10 e 15 g por planta. Os resultados indicaram que o fornecimento de nutrientes através do mineral zeólita adicionado ao substrato orgânico comprovou ser alternativa viável para a obtenção de porta-enxertos no sistema de produção em ambiente protegido. A adição de 6,4 g do concentrado zeolítico enriquecido com NPK aumentou significativamente a produção de matéria seca, área foliar, altura e diâmetro de caule. Este aumento foi de 37,5% em relação à testemunha que não recebeu o concentrado zeolítico. Houve aumentos nos teores e extração de N, P e K com o fornecimento da zeólita enriquecida. As leituras dos teores de clorofila relacionaram-se com os teores de N, indicando ser esta uma alternativa para o diagnóstico do estado nutricional para a cultura. |
