S18.201. aThe aim in this research work was the identification of polyploidy by means of stomata analysis and flow citometry, effects of poly and mixoploidy over anatomic and ultra structural traits on the following banana cultivars Malbut, Ouro, Lidi e Thong Dok Mak. Polyplodization was induced by colchicine treatment, in 0, 2,5; 7,5 e 12,5 mM concentrations, for 24 and 48 hours and by orizaline, in 0, 10, 30 e 50 ?M concentrations, for 4 and 7 days. Stomata analysis consisted of measurements of size and density by scanning electron microscopy (SEM). For the flow citometry analisys a small piece of the tissue was grinded and dyed with propidium iodide. Citogenetics analysis was performed using root tips pre treated with 8-HQ 500 mg.L-1 solution, during 3 hours, fixed in Carnoy and subsequently submitted to conventional squeezing method. Anatomic and ultra structural analysis was done in all the diploid, mixoploid and tetraploid plants of ?Lidi?, ?Ouro? and ?Tong Dok Mak?. Young leaf samples were traditionally prepared for transmission Electron Microscopy (TEM). Semi (>100nm) and ultra thin (<100nm) sessions were cut from the tissues. The semi thin sessions were analyzed by light microscope, measuring length and width of chlorophyll and air parenchymas, of superior and inferior epidermis, thickness of the leaves sessions and number of chloroplastids. TEM was used to analyze the ultra thin sessions to characterize the chloroplasts. The main results generated were, identification of 4 tetraploid and 6 mixoploid plants by three different techniques. Anatomical and ultra structural analysis made possible screening of diploids. Nevertheless, the distinction between tetraploid and mixoploids was not possible. aBanana aCultivares1 aPIO, L. A.
