Activity of the enzymes, alkaline phosphatase, acid phosphatase, glucose 6-phosphatase, phosphogiucoisomarase and sorbitol dehydrogenase were assayed in seminal plasma, sperm pack, ultrasonicate extract and ultrasonicate pack. The ultrasonicate extract was prepared by sonicating the semen pellet suspended in a suitable buffer (PBS/Tris) at 20 KCS using a vibronic ultrasonicator. The seminal plasma alkaline phosphatase activity was signifiicantly higher (P < 0.01) in both the breeda as compared to the other fractions. But a significant difference (P<0.01)in the seminal plasma activity was observed between the breeds, being higher in cross-bred. The soluble form of the enzyme was 65 to 70% and particulate bound was 30 to 35% in both the breeds. Acid phosphatase activity was higher in ultrasonicate extract compared to seminal plasma sperm pack and uirasonicate pack in both the breeds. Significant difference (P < 0.05) was observed in the seminal plasma activity, being higher in native breed. The soluble form of the enzyme was 80 to 82% and 18 to 20% was particulate bound. Glucose 6-phosphatase activity did not differ significantly between breeds. The soluble form of the enzyma was 45% and particulate bound was 55% it native breed and vice versa in cross-bred. Phosphoglucoisomerase activity in the seminal plasma was significantly lower (P <0.01) as compered to the sperm pack in both the breeds. About 92 to 95% of the enzyme was in the soluble form and 5 to 8% in particulate bound. Sorbitol dehydrogenase activity appearod to be low in seminal plasma as compared to sperm pack in both the breeds.
