V. aIdentificação de marcadores moleculares de microssatélites associados à resistência de genótipos de soja a Meloidogyne javanica.h[electronic resource] c2001 aTwenty loci of simple sequence repeats (SSR) were used to amplify the DNA of 120 F2 plants of the soyabean crosses BRS 133 (M. javanica-susceptible) x Coodetec 201 (resistant) and BRS 133 x PI 595099 (resistant). The loci are located close to some quantitative trait loci (QTLs) on the linkage group D1b+W and F. Each of the F2 (120) and F3 (120 families) generations and the parents (60 of each) were screened in a greenhouse for resistance to gall formation. The plants were grown in plastic tubes (4.5 cm in diameter x 19 cm length) filled with methyl bromide-fumigated soil amended with sand. The seedlings were individually inoculated with 3000 nematode eggs and evaluated by counting the root galls 30 days later. Analysis of variance indicated significant marker/nematode galls association for Satt 266 and Sat 133 (BRS133 x Coodetec 201) and HSP 176 (BRS 133 x PI 595099). This data corroborates the existence of QTLs conferring resistance to M. javanica located close to RFLP markers A725 on linkage-group D1b+W, and A806D and A186D/A757V on linkage-group F. However, the loci detected here did not explain much of the total phenotypic variation of the number of galls.
