Golden mosaic of common bean is caused by the Bean golden mosaic virus (BGMV). The disease is one of the greatest constraints on bean production in Latin America and causes significant yield losses. The RNAi concept was explored to silence the rep (AC1) viral gene and a transgenic bean line immune to BGMV upon inoculation at high pressure was previously generated. Identification of the transgene insert confirmed the presence of a single locus corresponding to two intact copies of the RNAi cassette in opposite orientation and three intact copies of the AtAhas gene. It is flanked by Phaseolus genomic sequences and interspersed by one nuclear and three chloroplastic genomic sequences. Southern analyses showed that the transgenes were structurally stable for eight self-pollinated generations and after backcrosses with a non transgenic commercial variety. Transgene expression analyses revealed similar levels of siRNA in leaves of transgenic plants cultivated under field conditions in three distinct regions. siRNA were also analyzed during seed development in common bean transgenic plants. siRNA signals were also detected in seeds, albeit at significantly lower levels than those observed in leaves, and could not be detected in seeds cooked during 10 min. This information is relevant to demonstrate that GM beans are free of siRNA signals after cooking and therefore suitable for human consumption. Additionally, characterization of the locus where the transgene was integrated in the common bean genome provides a valuable tool to trace this GM bean material in the field and in the market. O estado de Mato Grosso é um grande importador de frutas cítricas, o que, em função da grande distância dos polos produtores do Sudeste e do Nordeste, acarreta uma oferta de fruta de baixa qualidade para o consumo in natura e de alto preço para a população, em especial das cidades da região norte do estado. Esta região agrega condições ambientais e socioeconômicas promissoras para o desenvolvimento da cultura da limeira ácida ?Tahiti?. Este trabalho foi realizado com o objetivo de avaliar variáveis de crescimento vegetativo de limeira ácida ?Tahiti CNPMF 02? enxertada em 12 porta-enxertos. Os porta-enxertos foram gerados pelo Programa de Melhoramento Genético de Citros da Embrapa Mandioca e Fruticultura - PMG Citros e avaliados em Guarantã do Norte, MT. Os porta-enxertos utilizados foram: citrandarins ?Indio? [C. sunki (Hayata) hort ex Tanaka x Poncirus trifoliata (L.) Raf. ?English?] (CTRI) e ?San Diego? (C. sunki x P. trifoliata ?Swingle?) (CTRSD), citrumelo ?Swingle? (C. paradisi Macfad. x P. trifoliata) (CTSW), limoeiro ?Cravo?, clones ?Santa Cruz? e ?CNPMF-003?, tangerineira ?Sunki Tropical? (C. sunki) e os híbridos HTR - 069, TSKC x (LCR x TR) - 059, LVK x LCR - 038,TSKC x CTSW? 028,LRF x (LCR x TR) - 005,TSKC x TRFD 003 e 006. O delineamento experimental foi de blocos inteiramente casualizados, quatro repetições e doze tratamentos. As parcelas foram constituídas por cinco plantas. Com dois anos e seis meses de idade foram avaliados os seguintes caracteres: altura de planta, diâmetro do caule (10 cm acima e abaixo da linha de enxertia), diâmetro e volume da copa, fórmula V=2/3 x [(π x D/4) x H]. Observou-se que limoeiro ?Cravo?, clones ?Santa Cruz? e ?CNPMF-003?, a tangerineira ?Sunki Tropical?, o citrandarin ?Indio? e ?San Diego? foram superiores estatisticamente em relação ao crescimento vegetativo, apresentando uma altura média de 3,21 m e diâmetro 10,00 cm para a tangerineira ?Sunki Tropical?, que induziu maiores valores para altura e diâmetro de copa à limeira ácida à copa da limeira ácida ?Tahiti, ocorrendo o inverso com os híbridos HTR - 069, TSKC x CTSW?028, LRF x (LCR x TR) - 005, TSKC x TRFD 003 e 006, onde a altura média de plantas foi de 2,35 m e o diâmetro do enxerto foi de 8,39 cm para TSKC x TRFD 003 que obteve o menor valor dentre os demais.
