Zinc ions are effective inhibitors of respiratory chain of mitochondria; Mitochondrial fragments and reconstructed respiratory chain are lees sensitive to Zn2+. Zn2+ inhibits esterification of inorganic phosphate in intact mitochondrial and submitochondrial and fragments and also decreases the rate of respiration. Zinc ions do not inhibit ATPase. Inhibition of respiration in mitochondria, due to Zn2+ in the concentration 10-6_10-5 M, is localized between cytochromes b and c1; by the concentration of Zn 2.5-5.10-4 M and higher, inhibited are also NADH and succinate dehydrogenase parts of respiratory chain and cytochrome oxidase. Inhibition of respiration by zinc ions is removed by addition of chelating agents, SH-compounds and ATP. Ions of bivalent iron also reverse the Zn actions; competitive nature of relationships between Zn2+ and Fe2+ action on the respiration in mitochondria is demonstrated. |
