Coffee synthesizes two unique isoprenoid diterpenes, cafestol (C20H28O3) and kahweol (C20H26O3), ent-kaurene compounds produced through the mevalonate or mevalonate-independent pathway. These compounds are relevant to beverage quality, plant defense, human health and nutraceutical properties. We investigate the expression profile of genes in the early (CaHMGR, CaMVD, CaDXR and CaIDS) and final steps of isoprenoid diterpenes biosynthesis (Coffea arabica L. cytochrome P450s: CaCYP82C2, CaCYP71A25, CaCYP74A1 and CaCYP701A3) in two fruits development stages (90 and 210 days after flowering). We also observed total lipid, cafestol and kahweol accumulation in C. arabica grown using two crop systems (full sun and agroforestry) at IDR-Parana Londrina, PR, Brazil. Plants in full sun presented higher gene expression than plants grown in agroforestry systems, except for CaHMGR. Fruits showed higher levels of transcript activity 210 days after flowering, about 2.8 times more in the genes expressed during the first steps and 3.6 times in those in the later steps. Although lipid, cafestol and kahweol were higher in the full sun than the agroforestry system, only increased lipid content was statistically significant. Cultivation system plays an important role in the expression of the genes, particularly as regards light incidence. |

RESUMO: Por reunirem constituições genéticas de diferentes origens e níveis de melhoramento, os bancos de germoplasma podem constituir-se em ótimas fontes de genes para programas de melhoramento. Para tanto, é fundamental que o melhorista conheça profundamente o germoplasma disponível em relação à variabilidade e ao seu desempenho agronômico. O objetivo deste estudo foi estimar a variabilidade genética de 356 acessos do banco de germoplasma de mandioca da Embrapa Cerrados em relação a 27 descritores morfológicos, estabelecer quais desses evidenciam maior entropia e agrupar os acessos em função da sua dissimilaridade genética. No ano de 2004, 356 acessos de mandioca foram caracterizados por meio de 27 descritores morfológicos. Com base nos dados aferidos, foi estimado o nível de entropia dos caracteres, a dissimilaridade genética e, posteriormente, os acessos foram agrupados pelo método de agrupamento de Tocher. Os acessos de mandioca estudados foram divididos em 34 grupos distintos e evidenciam elevada variabilidade genética. Os caracteres com as maiores entropias foram: cor externa do caule, cor do pecíolo, forma do lóbulo central e cor da folha apical, e os com as menores entropias foram: hábito de crescimento do caule, floração, textura da epiderme da raiz e constrições da raiz. ABSTRACT: Germoplasm collections may constitute excellent sources of genes for plant breeding programs, once they gather genetic compositions from different origins and breeding levels. Therefore, it is essential for the plant breeders to gain profound knowledge of the available germoplasm in what concerns its variability and agronomical performance. The aim of the present work was to estimate the genetic variability of 356 cassava accessions from the Embrapa Cerrados germoplasm collection in what concerns 27 morphological descriptors, to determine which descriptors would display higher entropy and to group the accessions in terms of genetic dissimilarity. In the year 2004, 356 cassava accessions were characterized in terms of 27 morphological descriptors. Based on analyzed data, the entropy level of the characters and the genetic dissimilarity were estimated, and lately, the accessions were grouped by using the Tocher method. The cassava accessions which were analized were divided into 34 different groups and present high genetic variability. The characters which presented the highest entropy levels were: stem external colour, petiole colour, shape of the central lóbulo and colour of the apical leaf, whereas the characters presenting the lowest entropy levels were: stem growth habit, flowering, texture of the root epidermis and presence of constrictions on roots.
