RESUMO - Palmeiras (Arecaceae) são plantas com grande importância socioeconômica no mundo todo. Lagartas do gênero Automeris (Lepidoptera: Saturniidae) causam desfolhamento severo em palmeiras, bem como urticância quando em contato com a pele humana. Portanto, é extremamente importante a descoberta de inimigos naturais para tais pragas. O objetivo deste trabalho foi registrar e confirmar a ocorrência de uma nova espécie de baculovírus infectando Automeris liberia. Em maio de 2017, lagartas de A. liberia exibindo sintomas de infecção causada por baculovírus foram coletadas de dendezais na Tailândia, PA, Brasil. Em laboratório, um macerado das lagartas mortas foi oferecido a lagartas sadias de A. cinctistriga, reproduzindo os sintomas e levando-as à morte (postulado de Koch). Após purificação por ultracentrifugação em gradiente de sacarose, o material foi observado sob microscópio eletrônico de transmissão, revelando a presença de corpos de oclusão virais contendo múltiplos nucleocapsídeos, compatíveis com Alphabaculovirus (Baculoviridae). A detecção molecular por PCR utilizando primers dos genes polh, lef-8 e lef-9 confirmou que o isolado pertence ao gênero Alphabaculovirus. Estudos filogenéticos demonstraram que o mesmo se difere das espécies já conhecidas. Até onde os autores puderam investigar, este é o primeiro registro de um baculovírus isolado de espécies do gênero Automeris, ou associado a elas. O nome Automeris liberia nucleopolyhedrovirus (AuliNPV) foi proposto para este novo vírus.
ABSTRACT- Palms (Arecaceae) are plants with great socioeconomic importance worldwide. Caterpillars from the genus Automeris (Lepidoptera: Saturniidae) cause severe palm defoliation, as well as urtication when in contact with human skin. Thus, the discovery of natural enemies of such pests is very important. This work aims to record and confirm the occurrence of a novel baculovirus species infecting Automeris liberia. In May 2017, caterpillars of A. liberia from an African oil palm plantation in Tailândia, PA, Brazil exhibited symptoms of infection by viruses. In the laboratory, a macerated of dead caterpillars was offered to caterpillars of A. cinctistriga, reproducing the same symptoms and leading them to death (Koch’s postulates). After purification by ultracentrifugation on sucrose gradients, the sample was observed under the transmission electron microscope, which revealed the presence of viral occlusion bodies containing multiple nucleocapsids, compatible with Alphabaculovirus (Baculoviridae). Molecular detection by PCR with primers for genes polh, lef-8 and lef-9 confirmed that this isolate belongs to Alphabaculovirus genus whereas phylogenetic studies have shown that it is different from the already known species. As far as the authors were able to investigate, this is the first record of a baculovirus isolated from, or associated with, species of the genus Automeris.
