The nuclear gene Msh1 (MutS homologue 1), recently cloned in Arabidopsis (Abdelnoor et al., 2003, PNAS 100:5968-5973), is involved in regulation of the copy number of subgenomic mitochondrial DNA molecules created by ectopic recombination in a process known as substoichiometric shifting. The Arabidopsis genome contains several other MutS homologues, but MSH1 protein is the only one that is targeted to both mitochondria and chloroplast and the predicted computer-based structure of this protein is very similar to the E. coli MUTS. We have isolated and sequenced the full-length homologous Msh1 genes in soybean, rice, tomato, and common bean to better understand their structure and function in plants. The tomato and soybean genes were obtained based on EST sequences identified by similarity to the Arabidopsis Msh1 gene and by RT-PCR of total RNA from leaves. The rice Msh1 was identified by searching the rice BAC sequences database and the coding sequence was confirmed by RT-PCR. The bean Msh1 was isolated from a BAC library, using a sequence of the soybean Msh1 gene as probe. Similar to Arabidopsis Msh1, all of them are comprised of 22 exons with very well conserved size and splicing position. Phylogenetic analysis using ClustalW showed that these genes have an overall identity of about 60% at the protein level. Based on computer prediction analysis they are predicted to be targeted to mitochondria. Additionally, several other ESTs have been identified in other plant species (barley, wheat, potato, tobacco, sorghum, maize and medicago), and show high homology to the sequenced Msh1 genes. The alignment of all of these MSH1 proteins has allowed us to identify specific conserved domains that might be very important for their proper function in plant mitochondria. Resumo: Foram utilizados 2916 registros de nascimentos de cordeiros da raça Morada Nova, com o objetivo de avaliar o efeito da inclusão da covariância aditiva-materna na estimativa de parâmetros genéticos para peso ao nascimento. O modelo continha os efeitos de grupo contemporâneo (animais nascidos na mesma estação e ano, do mesmo tipo de nascimento e mesmo sexo, submetidos ao mesmo manejo), ordem de parto, efeito genético aditivo direto do animal, efeito genético aditivo materno e efeito de ambiente permanente da mãe. Foram realizadas duas análises, uma considerando a estimativa da covariância entre o efeito genético aditivo direto e materno e a outra considerando como zero esta covariância. Os modelos COM (com a inclusão da covariância) e SEM (sem a inclusão da covariância) foram comparados pelo teste de razão de verossimilhança em nível de 5% de probabilidade. Não houve diferença significativa entre os modelos. Entretanto, as estimativas de herdabilidades diretas para PN foram 0,24 e 0,18 para COM e SEM, respectivamente, e as herdabilidades maternas foram 0,16 e 0,08, seguindo a mesma ordem. Apesar de não haver diferença estatística entre os modelos, a inclusão da covariância entre os efeitos direto e materno, no modelo de análise para peso ao nascimento, revelou maiores valores para os componentes de variância e para os coeficientes de herdabilidade, o que mostra que sua inclusão permite o aumento da sensibilidade na estimativa dos parâmetros. [Effect of inclusion of direct-maternal covariance on the model to estimate genetic parameters for birth weight in Morada Nova sheep]. Abstract: Data from 2916 records of birth weight of lambs of Morada Nova breed were used with the aim of evaluating the inclusion of the effect of direct-maternal covariance in the estimate of genetic parameters for this trait. The model contained the effects of contemporary group (animals born on the same year and season, from the same birth type, and same sex, under same management), lambing order, direct additive genetic effect of the animal, maternal additive genetic effect and permanent maternal environmental effect. Two analysis were performed, one considering the estimative of the direct-maternal covariance and the other considering this covariance as zero. The model COM (with the covariance) and SEM (without covariance) were compared by likelihood ratio test with 5% of probability. There was no significant difference between the models. However, the direct heritabilities for birth weight were 0.24 and 0.18 according to models COM and SEM, respectively, and the maternal heritabilities were 0.16 and 0.08, following the same order. Despite there is no statistical difference between the models, the inclusion of the covariance between the direct and maternal effect in the model of analysis for birth weight revealed higher values for the components of variances and heritabilities indicating that its inclusion allows for increased sensitivity analysis of the parameter estimates.
