On slow-growing strain of Bradyrhizobium japonicum (USDA-110), one fast-grower Rhizobium fredii strain (USDA-205) and one Rhizobium sp. (AH-012) isolated from peanut nodules were inoculated on four species of legumes, namely soybean (Glycine max L. Merrill.) cv. Williams, cowpea (Vigna unguiculata ssp. cylindrica L.), mungbean (Vigna radiata L. Wilczek) and peanut (Arachis hypogaea L.). The strain USDA-110 nodulated all of the legumes except mungbean, USDA-205 nodulated the four species, and the strain AH-012 did it on cowpea and peanut. The B. japonicum strain fixed Nitrogen effectively on soybean and cowpea, the R. freddi strain fixed much more N2 on cowpea and mungbean than on the soybean variety tested, and the strain AH-012 showed nitrogenase activities only on peanut. The nodules of the slow-growing rhizobia (USDA-110) displayed a high Nitrate-reductase activities on soybean and cowpea, producing no nitrites at all on peanut; likewise, the fast-grower strain also reduced Nitrate under a high rates on cowpea. These data are significantly correlated to those for the Nitrogen fixed (+0.76). The uptake hydrogenases were expressed by bacteroids of the USDA-110 from the soybean and cowpea nodules (0.23 and 0.32 umol H2.h.-1. mg-1 protein), and the USDA-205 took up significant amounts of H2 on cowpea (0.17 uM H2.h-1.mg-1), though behaved as a Hup- strain on soybean and mungbean. This is the first report about H2 uptake by a R. fredii strain. |
