[Relations muscle/bone and muscle/fat 12th rib of sheep of different genetic groups].
Abstract: This study aimed to evaluate the influence of different genetic groups of sheep on the muscle/bone and muscle/fat 12th rib. We used 24 animals Santa Ines e 24 Ile de France x Santa Ines. The animals were reared under confinement until they reached slaughter weight (30, 35, 40 and 45 kg). After slaughter the carcasses were divided into two half carcasses and stored at -20°C. Were separated tissues: muscle, bone and fat, and estimated the ratio muscle/bone and muscle/fat. The design was completely randomized factorial 2 x 4. Had no influence on the muscle/bone ratio in the fraction of the 12th rib to the weights of animals slaughtered in Santa Ines, who at 30 kg had higher muscle/bone ratio (1.89) when compared with the Ile de France x Santa Inês (1.55). In the muscle/fat were not different slaughter weights. The slaughter weight of lambs of different genotypes did not provide differences in muscle/fat fraction of the 12th rib. To monitor the colonization of wheat roots by Azospirillum brasilense, we constructed several plasmids based on the pBBR1replicon expressing the gfp and gusA genes constitutively. Both genes were placed under control of the gentamycin resistance
gene promoter resulting in high levels of expression in Escherichia coli and A. brasilense. The constructed plasmids were stably maintained in A. brasilense strains even in the absence of selective pressure. The colonization of wheat plants grown under controlled conditions in sterilized vermiculite by A. brasilense strain FP2 (aSp7-derivative) transconjugants containing these plasmids was monitored. Bacteria expressing GFP were easily observed in fresh plant material by fluorescence microscopy. Cell aggregates and single bacteria were visualized on the surfaces of young root zones, such as roots hairs and lateral roots. Large cellular clumps were observed at the points of lateral root emergence or at intercellular spaces of root epidermal cells 30 days after inoculation. Although we failed to detected bacteria in internal cortical and xylem tissues of wheat
roots, the initial stage of endophytic colonization by A. brasilense may involve the sites detected in this work.
