Resumo: A osteocondrose (OC) é uma das principais causas de claudicação em suínos, afetando até 80% dos animais reprodutores e de produção, levando a perdas econômicas e reduzindo o bem estar. Com o objetivo de identificar genes e mecanismos genéticos envolvidos em estágios iniciais de OC em suínos, amostras da cartilagem articular distal do fêmur de 3 marrãs normais (controle) e 3 afetadas com OC latens foram coletadas, submetidas a análise histológica e extração total do RNA. As bibliotecas de RNA foram preparadas e sequenciadas em equipamento HiSeq 2500 (2x100pb). As sequências foram submetidas ao controle de qualidade e 98% delas foram mapeadas no genoma do suíno (Ensembl 84). A expressão diferencial foi obtida utilizando-se o EdgeR e a anotação funcional foi realizada com a ferramenta DAVID. Um total de 11.231 genes foi expresso na cartilagem do fêmur e 2.019 genes foram diferencialmente expressos, sendo 1.040 mais expressos e 979 menos expressos no grupo afetado com OC, incluindo genes codificantes, lincRNA e miRNAs. Considerando a análise funcional, diversos processos biológicos foram encontrados, tais como regulação da matriz extracelular, ossificação, ativação endotelial e estresse oxidativo. Novos mecanismos envolvidos na manifestação da OC latens foram evidenciados, contribuindo para o melhor entendimento desta condição em suínos e outras espécies de mamíferos que apresentam problemas locomotores como similares a osteocondrose. Abstract: Osteochondrosis (OC) is a major cause of lameness in sows and finisher pigs, affecting up to 80% of the animals, leading to economic losses and reducing the animal welfare. Aiming to identify genes and genetic mechanisms involved in the early stage of OC in pigs, samples of the distal femoral articular cartilage of 3 gilts (5 months of age) with OC latens and 3 normal gilts (control) were collected, submitted to histological analysis and total RNA extraction. RNA-Seq libraries were prepared and sequenced in HiSeq 2500 equipment (2x100pb). The reads were processed, cleaned and 98% were mapped to the pig genome (Ensembl 84). Differential expression (DE) was obtained using EdgeR and functional annotation was performed with the DAVID database. A total of 11.231 genes was expressed in femoral cartilage and 2.019 were DE, being 1.040 upregulated and 979 downregulated in the OC group, including coding genes, lincRNA and miRNAs. Considering the functional analysis, several biological processes were found, such as those related to regulation of extracellular matrix, ossification, endothelial activation and oxidative stress. We demonstrated novel pathways involved with OC latens contributing to the understanding of its pathophysiology in swine and other mammalian species. This work has as its objectives: a) to assess the geographical distribution and population structure of Amburana cearensis var. acreana; b) to calculate sustainable cutting rates, according to stipulated cutting cycles, and c) to simulate the projected recovery potential in volume based on the calculated cutting rate. It was used data from sustainable forest management plans, and the results will contribute for future decisions about its endangered condition. The results did not corroborate the information that Amburana cearensis var. acreana is endangered in Acre state. However the management sustainability will only be feasible if considered the ideal remaining population structure and the estimative of the optimal cutting rate according to the cutting cycle. |
