Anticarsia gemmatalis nuclear polyhedrosis virus (Ag-NPV) is an insect patogen, member of the Baculoviridae family. This group of virus contains a large, circular double-strand DNA genome at 7,5-10 x 10 daltons, that replicates in the nuclei of infected host cells. The infection leads to the formation of polyhedral inclusion bodies referred polyhedra (Summers & Smith, Virology, 84:390-402, 1978). Studies on the replication of baculovirus has been demonstrated the induction of DNA-dependent DNA polymerase (Miller et al., J. Virol., 40:305-308, 1981) and alpha-amanitin resistant DNA-dependent RNA polymerase (Fucns et al., J. Virol., 48:641-646, 1983) by Autographa californica nuclear polyedrosis virus in lepidoptera infected cells. This results suggest the presence of a viral RNA polymerase codified by the viral genone. In this communication we report a possible DNA-dependent RNA polymerase associated with Ag-NPV polyhedra. The enzyme activity was measured after activation the mixture contained polyhedra with sarkosyl or alpha-chymotrypsin. The enzyme was resistant to alpha-amanitin, and the addition of rifampicin, actinomycin D. ethidium bromide or neutral acriflavin, inhibit the transcription activity to some extent. The transcription products analysed by sucrose gradient centrifugation shows the presence of 3 RNA peaks. This results suggest a possible presence of RNA polymerase associated with Ag-NPV polyhedra. |
