Two specific primers were used to amplify possible lipoxygenase DNA fragments from root and nodule cDNAs of soybean plants. At least a major form of lipoxygenase expressed in the root was not expressed in the nodule. In order to infer putative functions of lipoxygenase present in the nodule, the PCR fragment from the nodule was cloned and sequenced resulting in a 356 bp sequence. The amino acid derived sequence of this cDNS clone, GmLOXN, when compared with others in the GenBank revealed to be a new form of a lipoxygenase gene. The homology of 69% of GmLOXN with vlxC, a lipoxygenase with possible role in temporary nitrogen storage, and the fact that this protein type accumulates in soybean in response to nitrogen fertilization, suggests the possibility of GmLOXN to be part of a gene involved in the temporary nitrogen storage in the nodules for such structures presenting high levels of nitrogen fixed. |
