Conteúdo:  Abstract: The objective of this investigation was to assess the use of a mutant form of the green fluorescence protein as a reporter gene in cassava. Sterile leaf explants were plated onto the primary somatic embryogenesis induction medium consisting of MS medium supplemented with 6 mg / l 2.4 - D (2, 4 - Dichlorophenoxyacetic acid), 2% sucrose and solidified with 0.25% Gelrite. After 20 days embryogenic callus was plated onto the secondary embryogenesis proliferation medium consisting of double strength MS medium and salts (pH 5.8) supplemented with 1 mg / l picloram, 2% sucrose and solidified with 0.25% Gelrite. Agrobacterium tumefaciens strain LBA4404 containig binary plasmid XC-1 blue encoding the green fluorescence protein, variant mgfp5, was used to transform secondary somatic embryogenic cells. Callus and embryos were examined after 7, 14, and 28 days following transformation for GFP activity using the Confocal microscope (Zeiss inverted microscope LSM 41000), fitted with a filter set suitable for excitation at 480nm and 509nm emission peak. In the transgenic cassava embryos GFP expression was found predominantly in the plasmalemma and nuclear membranes, and was generally excluded from the membranes of the vacuoles, plastids and other organelles within the cell. These results indicate that the GFP gene has potential as a reporter gene system for transformation experiments in cassava.
