Abstract: The distribution of the enzyme △5-3β-(3β-HSD) and 17-βhydroxysteroid dehydrogenases (17β-HSD) and NADH- and NADPH dehydrogenases was studied histochemically in the ovary of the nonpregnant (Day 5 of the estrous cycle) and early pregnant (Day 5)Tokara goat. Weak activities of NADH- and NADPH dehydrogenase reactions were demonstrated in the theca and granulosa cells of preantral follicles. In antral follicles these enzyme activities were very strong in the theca interna and moderate in the granulosa cells. These enzyme activities were also demonstrated in the theca externa of antral follicles, corpora lutea and germinal epithelia. There were no differences in the distribution and intensity of these two dehydrogenases between the ovary of the nonpregnant goat and the ovary of the early pregnant goat. The activity of 3βHSD was found in the corpus luteum where the enzyme activity was moderate and very slight activity was also found in the theca interna of the follicle over 5mm in diameter. These findings show that the corpus luteump plays an essential role in the synthesis of progesterone in the goat ovary and that the theca interna of antral follicles over 5mm in diameter can also contribute to the progesterone synthesis. The activity of 17β-HSD was mainly found in the granulosa cells of antral follicles. This enzyme activity in the granulosa cells of the early pregnant goat ovary was weaker than that in the nonpregnant goat ovary. These results suggest that the granulosa cells of antral follicles are capable of synthesizing estrogens in the goat ovary. None of 3β- and 17β-HSD activities were present in preantral follicles, suggesting that preantral follicles in the goat ovary are unlikely to contribute to steroidogenesis.
