The recent interest in the microbial conversion of sterol to C19 steroids was caused by the incresead demand for steroid drugs and the increased demand for steroid drugs and the shortage of diosgenin. Based upon studies on the pathway of sterol metabolism by certain bacteria methods for the selective cleavage of the 17-alkyl side chain of cholesterol and phytosterols were developed on a laboratory scale. These methods consist in the inhibition of the enzymas responsible for the primary attack of the steroid ring system. Structural modification of the substrates, fermentation in the presence of enzyme inhibitors, or mutagenic treatment were employed to achieve this goal. Since the microbial process has to compete witch chemical methods, higs, high conversion efficiencies in the presence of large substrate concetrations have to be obtained. Some of these process are used now an industrial scale for the production of 17-ketosteroids. |
