We investigated the hypothesis that excretion of organic acid is a mechanism of Al tolerance in wheat. Seedings of Al-tolerant wheat cultivars, Atlas 66 (AT) and Shirosanjyaku (SH), and Al-sensitive ones, Chikushikomugi (CK) and Saitama 27 (AS), were exposured to Al in experiments lasting from 40 minutes to 24 hours. Al induced differential pH changes in the solution. Al triggered 3-5 fold more malate in Al-tolerant cultivars than the Al-sensitive anes. Citrate in root exudates was not detected. The Al absorbed by roots of SH na CK within 2-hour tretment was mainly apoplasmically associated and could be removed from this compartment by an Al na chelator (citrate), as indicated by the lack of hematoxylin staiming of roots tips. The effect os Al on the metabolism of malate, synthesized via dark carbon fixation in roots demosntrated that Al triggered 14C excretion and malate to solution, mainly in SH. However, Al decreased malate concentrations in roots of both cultivars. These results suggest taht differential Al tolerant between SH and CK consited of the greater ability of the Al tolerant cultivar to excrete malic acid from roots and not and not due to its concentration in the roots. |
