O Brasil é o segundo maior produtor mundial de soja [Glycine max(L.) e, entre os principais fatores bióticos que contribuem para essa elevada produção destaca-se a fixação biológica do nitrogênio (FBN). O sucesso da FBN com a cultura no país resulta de pesquisas de seleção de estirpes de Bradyrhizobium compatíveis com as cultivares brasileiras, com alta eficiência de FBN e adaptadas às diferentes condições edafoclimáticas de cultivo. O presente trabalho teve como objetivo avaliar a variabilidade genética de cultivares de soja selecionadas para a maior eficiência na fixação biológica de nitrogênio quando inoculadas com estirpes comerciais de Bradyrhizobium utilizadas no Brasil e para alto teor de proteína nos grãos. Foram avaliadas 191 cultivares desenvolvidas por instituições públicas e privadas. O DNA genômico foi extraído de folhas jovens (estágio V3) e amplificado com 22 marcadores microssatélites (SSR) (Satt050, Satt385, Satt455, Sat_270, Satt251, Satt509, Satt416, Satt180, Satt338, Satt578, Satt202, Satt277, Satt357, Satt531, Satt192, Satt239, Satt571, Satt406, Satt567, Sat_084, Satt159, Satt478). A diversidade genética foi calculada de acordo com a expressão: 1-∑ Pij 2, onde Pij é a frequência do alelo i para o lócus h considerando todos os alelos no lócus i. A matriz de dissimilaridade foi utilizada para a obtenção dos agrupamentos de genótipos baseados nas distâncias entre cada par de genótipos, utilizando UPGMA (unweighted pair-group method), implementada no programa DARwin 5.0. Entre os 22 marcadores SSR avaliados, somente dois (Satt455 e Satt578) não apresentaram polimorfismo. O número total de alelos foi de 102, variando de 2 a 12, com uma média de 5,1 alelos por lócus. A média do número efetivo de alelos (Ae) foi de 4,28, variando de 1,46 (Satt567) a 10,6 (Satt416). O índice de polimorfismo variou de 0,2 (Satt567) a 0,8 (Satt406), com uma média de 0,48. O dendrograma gerado pelo método Neighbor-Joining mostrou a relação genética entre as 191 cultivares de soja e separou-as em dois grupos distintos. Os resultados obtidos no presente estudo possibilitaram a identificação dos marcadores SSR mais informativos e a existência de alta variabilidade entre as 191 cultivares. Essas informações poderão ser úteis em programas de melhoramento genético de soja. Blast fungus (Magnaporthe oryzae B.C. Couch) is an imminent threat to global food security because it causes serious yield losses in rice (Oryza sativa L.) and wheat (Triticum aestivum L.). The investigation of infection processes in resistant and susceptible varieties, as well as the cellular responses involved in resistance, can help us to better understand the process of interaction of the M. oryzae-Poaceae pathosystems. Thus, the objectives of this study were to evaluate the processes of pre-and post-infection of M. oryzae in leaves of wheat varieties with diferent levels of resistance. The percentage of germinated conidia, appressorium formed, tissue penetration and colonization, and the reaction of leaf tissue to infection were evaluated. A decrease was observed in the percentage of germinated conidia, appressorium formation, tissue penetration and colonization, especially in the tissues of resistant varieties, in addition to an increase in the plant?s response to infection, with cell wall reinforcement, cell death, and autofuorescent cytoplasm aggregation. Nevertheless, our data produced a diferent temporal perspective regarding the expression of the known types of resistance. We found that, for a single genotype, recognition can start as early as 6 h after inoculation and continue to evolve until very late during the infection cycle, culminating in cell death. The combined and overlapping pre- and post-haustorial resistance mechanisms were sufcient to prevent disease symptoms, with a few punctual lesions observed in one of the resistant varieties (BR 18) and no visible symptoms in the other two (Ônix or BRS229) as opposed to susceptible variety.
