Abstract: The aim was to verify the correlation between follicular population count, superovulatory response and the recovery of viable structures in the in vivo production of sheep embryos. Twenty-five nulliparous Santa Inês ewes (11.9 ± 1.1 months old, body score of 2.8 ± 0.3) were superovulated using the Day 0 protocol concept. For previous wave synchonization, intravaginal progestagen sponges (Progespon®, Zoetis, Campinas-SP, Brazil) were kept for 6 days and on Day 5, 300 IU eCG(Novormon®, Schering Plough, São Paulo, Brazil) and 0.24mg cloprostenol (Estron®, Tecnopec, São Paulo, Brazil) were given. Thirty-six hours after sponge removal, 25 µg licerelin was administered.The superovulation started 80 hours after sponge removal by the use of 200 mg of FSH/ per ewe (Folltropin-V ®, Bioniche Animal Health, Ontario, Canada) in six declining doses, every 12 hours (50/50, 30/30, 20/20 mg). At the first FSH dose, a new sponge (Progespon®, Zoetis, Campinas-SP, Brazil) was inserted and removed at the time of the fifth dose. At the last FSH dose, 0.24 mg of cloprostenol (Estron®, Agener Union, São Paulo, Brazil) and, 24 hours later, 25 µg of lecirelin (Gestran Plus®, Tecnopec, São Paulo-SP, Brazil) were administered. Ewes were mated every twelve hours from the last FSH dose to the end of estrus. An ultrasound equipment (Sonoscape S6®, SonoScape, Shenzhen, China) coupled to a 7,5 MHz linear transducer, by transrectal via was used for the quantification of the follicular population (PF) in two moments: at the beginning of the estrus synchronization (PFESTRUSBASE) and at the time of the first FSH dose (PFFSH-1). Embryo collections were carried out by surgical method, six to seven days after mating, and the viable structures (VS) quantified. The number of CLs (NCLs) was determined by laparoscopy previously to the embryo recovery. The PFESTRUSBASE and PFFSH-1 variables were compared with the NCLs and EV through the Pearson's correlation coefficient and Simple Linear Regression Analysis. For all tests, P < 0.05 was considered as statistically significant. An average of 7.5 ± 4.8 CLs and 4.0 ± 3.5 viable structures were obtained per donor. Significant correlations, medians and low, were found between: PFESTRUSBASE and NCLs (r = 0.45; r2 = 0.17; P < 0.05); PFFSH-1 and NCLs (r = 0.41; r2 = 0.13; P < 0.05); PFESTRUSBASE and VS (r = 0.55; r2 = 0.27; P < 0.05) and PFFSH-1 and VS (r = 0.41; r2 = 0.13; P < 0.05). In conclusion, there is a median correlation between follicular population observed by ultrasonography and viable recovered structures after superovulation protocol. Therefore, this tool is not indicated as a screening tool, alone, in the selection of Santa Inês sheep embryo donors. [População folicular na estimação da resposta superovulatória em ovelhas da raça Santa Inês]. Resumo: Objetivou-se verificar a correlação entre a população folicular na resposta superovulatória e na recuperação de estruturas viáveis em programas de produção in vivo de embriões ovinos. Vinte e cinco ovelhas nulíparas da raça Santa Inês (11,9 ± 1,1 meses, ECC: 2,8 ± 0,3) foram superovuladas utilizando-se o cio base para início do protocolo (D0). Para sincronização prévia da emergência folicular, manteve-se por seis dias um implante vaginal contendo 60 mg de acetato de medroxiprogesterona (Progespon®, Zoetis, Campinas-SP, Brasil), 24 horas antes da remoção do implante aplicou-se 300 UI de eCG (Novormon®, Schering Plough, São Paulo, Brasil) e 0,24mg de cloprostenol sódico (Estron®, Tecnopec, São Paulo, Brasil). Trinta e seis horas após a remoção da esponja, os animais receberam 25 ?g de lecirelina (Gestran Plus®, Tecnopec, São Paulo-SP, Brasil). A superovulação iniciou 80 horas após a retirada da esponja. Utilizou-se 200 mg de FSH/por animal (Folltropin®, União Química Farmacêutica Nacional S/A, São Paulo, Brasil) em seis doses decrescentes a cada 12 horas (50/50, 30/30, 20/20 mg). Na última dose de FSH, aplicou-se 0,24 mg de cloprostenol (Estron®, Agener União, São Paulo, Brasil) e, 24 horas após, 0,025 mg de lecirelina (Gestran Plus®, Tecnopec, São Paulo-SP, Brasil). Em paralelo, na primeira dose de FSH uma nova esponja (Progespon®, Zoetis, Campinas-SP, Brasil) foi inserida e removida na penúltima dose deste hormônio. Após a última dose de FSH realizou-se monta natural controlada a cada 12 horas. Utilizouse um aparelho de ultrassom (Sonoscape S6®, SonoScape, Shenzhen, China) acoplado a um transdutor linear de 7,5MHz, por via transretal, para a quantificação da população folicular (PF) em dois momentos: no início do protocolo de sincronização do estro base (PFESTROBASE) e no momento da aplicação da primeira dose de FSH (PFFSH-1). As coletas foram realizadas por via cirúrgica, seis a sete dias após as coberturas, e as estruturas viáveis (EV) quantificadas. O número de CLs (NCLs) foi determinado por laparoscopia no dia da recuperação embrionária. As variáveis PFESTROBASE e PFFSH-1 foram comparadas com o NCLs e EV por meio do Índice de Correlação de Person e pela Análise de Regressão Linear Simples. Para todos os testes, considerou-se 5% de significância. Foram obtidos 7,5 ± 4,8 corpos lúteos e 4,0 ± 3,5 estruturas viáveis por doadora. Foram encontradas correlações significativas, medianas e baixas, entre o PFESTROBASE e NCLs (r=0,45; r2=0,17; p<0,05); PFFSH-1 e NCLs (r=0,41; r2=0,13; p<0,05); PFESTROBASE e EV (r=0,55; r2=0,27; p<0,05) e também entre o PFFSH-1 e EV (r=0,41; r2=0,13; p<0,05). Em conclusão, existe uma correlação mediana entre população folicular observada por ultrassonografia e estruturas viáveis recuperadas após protocolo de superovulação. Assim, esta metodologia não é indicada como ferramenta de triagem isolada na seleção de ovelhas doadoras de embriões da raça Santa Inês. For previous wave synchonization, intravaginal progestagen sponges (Progespon®, Zoetis, Campinas-SP, Brazil) were kept for 6 days and on Day 5, 300 IU eCG(Novormon®, Schering Plough, São Paulo, Brazil) and 0.24mg cloprostenol (Estron®, Tecnopec, São Paulo, Brazil) were given. Thirty-six hours after sponge removal, 25 µg licerelin was administered.The superovulation started 80 hours after sponge removal by the use of 200 mg of FSH/ per ewe (Folltropin-V ®, Bioniche Animal Health, Ontario, Canada) in six declining doses, every 12 hours (50/50, 30/30, 20/20 mg). At the first FSH dose, a new sponge (Progespon®, Zoetis, Campinas-SP, Brazil) was inserted and removed at the time of the fifth dose. At the last FSH dose, 0.24 mg of cloprostenol (Estron®, Agener Union, São Paulo, Brazil) and, 24 hours later, 25 µg of lecirelin (Gestran Plus®, Tecnopec, São Paulo-SP, Brazil) were administered. Ewes were mated every twelve hours from the last FSH dose to the end of estrus. An ultrasound equipment (Sonoscape S6®, SonoScape, Shenzhen, China) coupled to a 7,5 MHz linear transducer, by transrectal via was used for the quantification of the follicular population (PF) in two moments: at the beginning of the estrus synchronization (PFESTRUSBASE) and at the time of the first FSH dose (PFFSH-1). Embryo collections were carried out by surgical method, six to seven days after mating, and the viable structures (VS) quantified. The number of CLs (NCLs) was determined by laparoscopy previously to the embryo recovery. The PFESTRUSBASE and PFFSH-1 variables were compared with the NCLs and EV through the Pearson's correlation coefficient and Simple Linear Regression Analysis. For all tests, P < 0.05 was considered as statistically significant. An average of 7.5 ± 4.8 CLs and 4.0 ± 3.5 viable structures were obtained per donor. Significant correlations, medians and low, were found between: PFESTRUSBASE and NCLs (r = 0.45; r2 = 0.17; P < 0.05); PFFSH-1 and NCLs (r = 0.41; r2 = 0.13; P < 0.05); PFESTRUSBASE and VS (r = 0.55; r2 = 0.27; P < 0.05) and PFFSH-1 and VS (r = 0.41; r2 = 0.13; P < 0.05). In conclusion, there is a median correlation between follicular population observed by ultrasonography and viable recovered structures after superovulation protocol. Therefore, this tool is not indicated as a screening tool, alone, in the selection of Santa Inês sheep embryo donors. [População folicular na estimação da resposta superovulatória em ovelhas da raça Santa Inês]. Resumo: Objetivou-se verificar a correlação entre a população folicular na resposta superovulatória e na recuperação de estruturas viáveis em programas de produção in vivo de embriões ovinos. Vinte e cinco ovelhas nulíparas da raça Santa Inês (11,9 ± 1,1 meses, ECC: 2,8 ± 0,3) foram superovuladas utilizando-se o cio base para início do protocolo (D0). Para sincronização prévia da emergência folicular, manteve-se por seis dias um implante vaginal contendo 60 mg de acetato de medroxiprogesterona (Progespon®, Zoetis, Campinas-SP, Brasil), 24 horas antes da remoção do implante aplicou-se 300 UI de eCG (Novormon®, Schering Plough, São Paulo, Brasil) e 0,24mg de cloprostenol sódico (Estron®, Tecnopec, São Paulo, Brasil). Trinta e seis horas após a remoção da esponja, os animais receberam 25 ?g de lecirelina (Gestran Plus®, Tecnopec, São Paulo-SP, Brasil). A superovulação iniciou 80 horas após a retirada da esponja. Utilizou-se 200 mg de FSH/por animal (Folltropin®, União Química Farmacêutica Nacional S/A, São Paulo, Brasil) em seis doses decrescentes a cada 12 horas (50/50, 30/30, 20/20 mg). Na última dose de FSH, aplicou-se 0,24 mg de cloprostenol (Estron®, Agener União, São Paulo, Brasil) e, 24 horas após, 0,025 mg de lecirelina (Gestran Plus®, Tecnopec, São Paulo-SP, Brasil). Em paralelo, na primeira dose de FSH uma nova esponja (Progespon®, Zoetis, Campinas-SP, Brasil) foi inserida e removida na penúltima dose deste hormônio. Após a última dose de FSH realizou-se monta natural controlada a cada 12 horas. Utilizouse um aparelho de ultrassom (Sonoscape S6®, SonoScape, Shenzhen, China) acoplado a um transdutor linear de 7,5MHz, por via transretal, para a quantificação da população folicular (PF) em dois momentos: no início do protocolo de sincronização do estro base (PFESTROBASE) e no momento da aplicação da primeira dose de FSH (PFFSH-1). As coletas foram realizadas por via cirúrgica, seis a sete dias após as coberturas, e as estruturas viáveis (EV) quantificadas. O número de CLs (NCLs) foi determinado por laparoscopia no dia da recuperação embrionária. As variáveis PFESTROBASE e PFFSH-1 foram comparadas com o NCLs e EV por meio do Índice de Correlação de Person e pela Análise de Regressão Linear Simples. Para todos os testes, considerou-se 5% de significância. Foram obtidos 7,5 ± 4,8 corpos lúteos e 4,0 ± 3,5 estruturas viáveis por doadora. Foram encontradas correlações significativas, medianas e baixas, entre o PFESTROBASE e NCLs (r=0,45; r2=0,17; p<0,05); PFFSH-1 e NCLs (r=0,41; r2=0,13; p<0,05); PFESTROBASE e EV (r=0,55; r2=0,27; p<0,05) e também entre o PFFSH-1 e EV (r=0,41; r2=0,13; p<0,05). Em conclusão, existe uma correlação mediana entre população folicular observada por ultrassonografia e estruturas viáveis recuperadas após protocolo de superovulação. Assim, esta metodologia não é indicada como ferramenta de triagem isolada na seleção de ovelhas doadoras de embriões da raça Santa Inês.
