Abstract: Since the first production of transgenic plants by particle bombardment in 1996 the method has been constantly improved and new methods were developed. The luciferase selection method used for the production of the first transgenics had as disadvantage that only a fraction of the tissue was transgenic. The luciferase selection method was refined leading to complete transgenic cultures. Furthermore, transgenic plants have been produced via combination of luciferase selection with electroporation and via combination of kanamycin and hygromycin selection with Agrobacterium tumefaciens. The transformation methods will be presented and compared with each other with respect to time and labour required using a model. Also the effect of some improved methods of regeneration of plants from FEC are presented using this model. Furthermore, the morphology of the transgenic will be compared with control regenerants and control plants. |
