Bacillus thuringiensis (Bt) toxins are effective in controlling insect pests either through the spraying of products or when expressed in transgenic crops. The discovery of endophytic Bt strains opened new perspectives for studies aimed at the control of sap-sucking insects, such as the Asian citrus psyllid Diaphorina citri Kuwayama (Hemiptera: Liviidae), a vector of "Candidatus Liberibacter spp.," associated with citrus huanglongbing (HLB). In this study, translocation of endophytic Bt strains in citrus seedlings inoculated with Bt suspension delivered by soil-drench, and their systemic pathogenicity to D. citri nymphs were investigated. The pathogenicity of three wild-type Bt strains against D. citri third-instar nymphs was demonstrated. Among the 10 recombinant strains tested (each of them harboring a single cry or cyt gene), 3 can be highlighted, causing 42%-77% and 66%-90% nymphal mortality at 2 and 5 d after inoculation, respectively. The isolation of Bt cells from young citrus shoots and dead nymphs, and PCR performed with specific primers, confirmed the involvement of the Bt strains in the psyllid mortality. This is the first report showing the translocation of Bt strains from citrus seedling roots to shoots and their potential to control D. citri nymphs that fed on these soil-drench inoculated seedlings. The Bt strains that caused the highest mortality rates have the potential to be used as bioinsecticides to control D. citri and the identified genes can be used for the production of transgenic Bt citrus. A grevílea é uma espécie natural da Austrália. Foi introduzida no Brasil no final do século XIX para o sombreamento de cafezais sendo, também, usada para ornamentação, produção de serapilheira, quebra-ventos e madeira para fins diversos. Esse estudo objetivou estimar a produção de madeira e estabelecer fontes de sementes melhoradas para plantios comerciais no estado do Paraná. Para tanto, foi instalado em Ponta Grossa-PR um teste de progênie com dezoito procedências australianas da espécie, contendo 59 progênies. Para servir de comparação com as progênies importadas, foi utilizado um material comercial disponível no país. O teste foi instalado utilizando o
delineamento de blocos, ao acaso com parcelas lineares de cinco plantas, distribuídas aleatoriamente. Foram coletados dados referentes à altura total e ao DAP, para a avaliação do volume cilíndrico. Essa variável foi submetida à análise de variância para determinar o efeito de progênies e sítio no desempenho da espécie. Os resultados possibilitaram as seguintes conclusões: 1) As progênies australianas são em média 71 % superiores em produção volumétrica de madeira, quando comparadas com testemunha; 2) a melhor progênie e as doze melhores progênies australianas, foram 140 % e 119 % respetivamente superiores em produção volumétrica de madeira, quando comparadas com o material comercial; 3) a grevílea é altamente sensível à qualidade do sítio e as sementes disponíveis na região oriundas das primeiras introduções, apresentaram baixa qualidade genética, justificando a importação de novas progênies mais produtivas da espécie.
The Grevillea robusta Cunn is an Australian species and it was introduced in Brazil in the end of XIX century for shadow plantation of coffee. It is used to ornamentation, mulch and different wood utilities. The purpose of this study was to identify genetically superior individuais of wood volume production in order to assure good material for future seed orchard in the state of Paraná. The genetic material was based on 60 half-sib progenies from eighteen Australians provenance and one commercial controI. Progenies were evaluated in randomized blocks with seven replications, five plants per plot. Higher and diameter were colected and the volume was evaluated by the analysis of variance to determine the effect of progenies and ambiental environrnent on the wood production. The results concluded that: 1) The Australians progenies were 71 % more productive then commercial contral in volume of wood. 2) The best progenie and the twelve more productive progenies were 140 % and 11 9 % respectively more productive then commercial control in volume of wood. 3) Different enviranments interfere significantly in the average growth and the firth introduction of Grevillea were with low genetic quality and the importation of new provenance and progenies is justified.
