Importance of tissue culture in plant virology has been rightly emphasized in earlier reviews by raychaudhuri (1966) and Kassanis (1967). Plant Virologist's efforts have been to develop a system to test the viruses under precisely controlled conditions, a boon enjoyed by animal virus workers for a long time due to amicable animal tissue culture system. Not that plant virus systems are not available or that these don't get infected, but because their infection without injury has eluded the worker so far. Tobacco tissue cultures have been infected with tobacco mosaic virus by pricking the cultures with fine needles previously dipped in the inoculum (Kassanis et al, 1958). This method was also found useful in inoculating potato virus X (Moskovets et al, 1971). Susceptibility of cultures varies according to age: 6-8 week old tissue of tobacco were found to be most susceptible to PVX (Moskovets et al, 1971) and all the cells in a culture don't get uniformly infected (Hildebrandt and Riker, 1958). |
