The araucaria forest (mixed ombrophilous forest with Araucaria angustifolia Bertoloni or the subtropical evergreen seasonal conifer forest) is one of the most important biomes that originally covered a large part of Southerm Brazil. With human population growth and expansion of agriculture, this forest was cut extensively and currently occupies less than 3% of its original area. large continuous forests wer also fragmented during this process and A. angustifolia populations are scattered in islands. This deforestation process probably ressulted in genetic erosion of A. angustifolia. Knowledge of diversity and distribution of genes within species and their local populations is an essential tool for the management and conservation of that species and biome. The aim of the present study was to analyse the genetic variability and differentiation among five populations of A. angustifolia through RAPD markers. Eleven DNA extraction protocols were tested in a factorial design, including use of different types of deproteinizing and antioxidant agents and detergents. Good quality DNA from buds of adult A. angustifolia trees was obtained only with a complex treatment which included addition of sarcosil to the extraction buffer, additional steps of phenol treatments, chloroform-isoamyl alcohol and low concentration-NaCl 1% CTAB reprecipitation. Other methods wer evalueted in attempts to simplify the process, increase yield and quality of DNA, and to decrease costs. The trial includes the determination of the leaf maturity stage from which good quality DNA could be obtained in large quantity. Quantity of DNA varied in the range of 20-1000 ng/ul, with higher values for buds and terminal needles. One hundred twenty random ten-base primers are being evaluated in five provenances, using bulking strategy, to determinate polymorphic and reproducible RAPD markers. Analysis of genetic structure in remnants of araucaria forests with the use of RAPD markers and gene flow between islands with the use of microsatellite markers will follow.
