Cotyledon explants were cultured for up to 8 wk on Gresshoff-Doy shoot induction medium supplemented with benzylaminopurine [benzyladenine] and NAA, to which was added ABA at concn. of 10-8, 10-7 or 10-6 M. Best results were obtained with 10-7 M ABA for 4 wk. This treatment doubled the morphogenic area of cotyledons and increased the fresh wt. of cotyledon explants by 40-45%. Comparison of 2 Texas, USA, seed sources showed that response to ABA was related to explant seed source and was not increased by prolonged exposure. Compared to controls, shoot number was increased by 31% and 56% with 2 and 4 wk respectively of exposure to 10-7 M ABA. |
