RESUMO: O objetivo do presente trabalho foi avaliar o efeito de concentrações de ácido indolbutírico (AIB) no enraizamento in vitro de brotações juvenis de Corymbia citriodora, e formas de vedação na aclimatização de plântulas micropropagadas. Para o enraizamento, foram colocadas brotações em meio de cultura com quatro concentrações de AIB (0; 1,0; 2,0 e 4,0 mg L-1). Na pré-aclimatização, plântulas germinadas in vitro e isentas de contaminação foram transplantadas para copos de polietileno contendo substrato. Os copos foram embalados em sacos plásticos por 10 dias e mantidos em sala de cultura. Posteriormente, foram aplicados três tratamentos: ausência de cobertura; saco plástico intacto e saco plástico perfurado. Na aclimatização, manteve-se o histórico dos tratamentos da pré-aclimatização. As plântulas foram transplantadas para tubetes contendo substrato e mantidas inicialmente em casa de vegetação e depois em casa de sombra. Os resultados demonstram que as concentrações de 1,0; 2,0 e 4,0 mg L-1 de AIB foram eficientes no enraizamento de explantes juvenis de C. citriodora, sendo que a maior concentração utilizada proporcionou o maior número médio de raízes. Recomenda-se a ausência de cobertura na pré-aclimatização para maior taxa de sobrevivência, altura e massa seca das mudas na etapa de aclimatização. TITLE: In vitro rooting and acclimatization of Corymbia citriodora seedlings ABSTRACT: The objective of this study was to evaluate the effect of indolbutyric acid (IBA) concentrations on in vitro rooting of Corymbia citriodora juvenile shoots, and sealing forms on the acclimatization of micropropagated seedlings. For rooting, shoots were placed in a culture medium with four concentrations of IBA (0, 1.0, 2.0, and 4.0 mg L-1). In pre-acclimatization, seedlings germinated in vitro and without contamination were transplanted into polyethylene cups containing substrate. The cups were packed in plastic bags for 10 days and kept in a culture room. Subsequently, three treatments were applied: no coverage; intact plastic bag, and perforated plastic bag. In acclimatization, the history of pre-acclimatization treatments was maintained. The seedlings were transplanted into tubes containing substrate and kept initially in a greenhouse and then in a shade house. The results demonstrate that concentrations of 1.0; 2.0 and 4.0 mg L-1 of IBA were efficient in the rooting of juvenile explants of C. citriodora, and the highest concentration used provided the highest average number of roots. The absence of cover in pre-acclimatization is recommended for greater survival rate, height, and dry mass of seedlings in the acclimatization stage. The project concerns an experiment in recultivating a fallow rubber plantation of 19 ha by establishing mixed plantations of selected crops, mainly tree. In order to achieve sustainability, different crop combinations and different strategies for management of the spontaneous vegetation will be tested, and the crop systems will be stabilized by inoculation the plants with spores of mycorrhizal fungi. The experimental area was divided into five blocks with eighteen plots each to test four mixed cultivation systems, compared to four conventional monoculture systems. Fallow plots will be analyzed for reference. The scientific research is focused on the ecological role of mycorrhizal fungi in the field trial, analysis of the structural and functional traits of the structural and functional traits of the spontaneous vegetation due to management, and identification and control of plant diseases. In addition, economic and acceptance studies will be conducted to find out whether farmers are willing to apply the tested cultivation systems. In the 19 ha plantation, intend to test the three following ways of stabilizing crops using different variants and to analyze the crop systems on a scale that is close to practical conditions: 1. inoculation of the plants with spores of mycorrhizal fungi; 2. testing of different mixed cultivation systems; and experiment on management of the spontaneous vegetaion in the crop systems to improve the competitive conditions for the planted crops. |
