Leprosis is a complex pathosystem that comprises a virus that does not spread systemically within its hosts, an unusual mite vector, Brevipalpus sp., and an ever-growing list of hosts. Although reported more than one century ago, the increasing importance and losses caused by the disease to the citrus industry in the Americas have led to the intensification of the studies on leprosis. In the last decade, most of what was accepted about CiLV-C and its interactions with the host and the vector was shown to be inaccurate. Our data bring novel information on the virus-vector relationship (believed to be propagative and now shown to be of circulative manner) as well as the virus-host interaction (initially thought to be a HR-type of resistance and now shown to be a typical compatible host-virus interaction). In order to understand the mechanisms involved in the response of susceptible and resistant citrus genotypes to the virus, we performed a genome-wide study using a customized microarray chip containing ~32,000 unigenes from Citrus sinensis. Analysis was done to identify the most representative metabolic pathways using Arabidopsis thaliana Gene Ontology as basis. The differentially expressed genes were compared to Arabidopsis thaliana submitted to biotic stress microarray experiments with the software Genevestigator. From these comparisons, 50 genes were selected and are being validated by RT-qPCR. |
