In order to better understand the organization and support sugarcane breeding programs we are beginning a construction of a physical map using BAC-FISH. We began the study with six clones BACs available at Alellyx S.A. These BACs were pre-selected for specific promoters search project. Filters of the sugarcane BAC library and the clones were obtained from Clemson University Genomics Institutes (CUGI, Clemson, South Carolina). This BAC library was constructed from genomic DNA of the hybrid 'R570. BAC DNA was shotgun sub cloned and was sequenced using BigDye system and ABI Prism 3730 DNA Sequencer (Applied Biosystems). Sequence homology was determined by BLAST searches of the nucleotide sequence database at the National Center for Biotechnology Information (NCBI). We analyzed the BLAST results and we could classify these six BACs when compared with sorghum genome database. |
