The soybean looper (Chrysodeixis includens) is an agricultural pest that affects several crops. Traditional control measures often involve chemical insecticides, which are harmful and can lead to the selection of resistant insect. Alternatively, natural products based on the baculovirus Chrysodeixis includens nucleopolyhedrovirus (ChinNPV) are both safer and widely used in Brazil. Baculoviruses are enveloped viruses with circular double-stranded DNA, enclosed within proteinaceous crystals known as occlusion bodies (OBs). This study aims to characterize two ChinNPV isolates from the EMBRAPA/CENARGEN Invertebrate Virus Collection: the reference isolate CNPSo168 (C168) and the Tabatinga isolate (Tb), to identify the isolate that is fastest to kill, most lethal, and most UV-resistant for effective field application. The original isolates were amplified in caterpillars, semi-purified, and quantified. Viral DNA from the amplified isolates was extracted, analyzed on agarose gel, and subjected to lethal dose assays using the original stock. Survival curves and UV exposure tests at a 1x10^7 OB/mL dilution for 10 days were also conducted. DNA analysis of the amplified viruses revealed bands corresponding to the ChinNPV genome and RNA segments typical of cypoviruses, confirming co-infection after amplification. This co-infection was verified by qPCR for a cypovirus specific gene. Cypovirus co-infection can significantly reduce the number of baculoviral occlusion bodies (OBs) and decrease insect lethality, posing a major challenge for baculovirus manufacturing. Consequently, we reverted to the original stock for further testing. In the lethal dose assay with the original samples, the Tb isolate showed a mortality rate of 68%, while the C168 isolate showed 81% at a 1x10^6 dilution. During UV exposure tests, C168 maintained a lethality of 13%, and Tb had a lethality of 12.5%. The complete genomes of these isolates, including those with cypovirus co-infection, will be sequenced.
