Abstract: Due to sensitivity to drought of potato, associated with the expansion of this crop to drier regions, the demand for phenotyping of tolerant genotypes grows. To evaluate the response in behaviour of plants to this factor, the ideal condition for selecting material must be defined beforehand. Thus, the objective of this experiment was to evaluate the behaviour of potato plants under water deficit by osmotic induction in periods with different irradiance levels. Six potato genotypes were used: Agata, C2397-03, BRS Clara, C2406-03, Cota and C2364-05–02. To subject the genotypes to water deficit, polyethylene glycol (PEG) was added to the nutrient solution; after this, photosynthetic variables and shoot temperature were evaluated. During the last gas exchange evaluation, leaf and tuber tissue samples were collected to quantify levels of total soluble sugars, sucrose, proline and the osmotic potential. At the end of the cycle, the tuber number and percentage of tuber dry weight were evaluated. The entire experiment was conducted in two periods (April/June and September/November) with different irradiance levels. Thus, it is clear that in the spring/summer period, when there is more irradiance, the stress applied was more intense on the genotypes evaluated. Furthermore, according to the tuber production results, in the second period, the plants better expressed their productive potential, in addition to a stronger correlation between the measured variables. The potential for tolerance to water deficit is related to the maintenance of leaf gas exchange and the increase in proline and sugar content in tubers.
