Abstract: Cassava explants of the variety SS4 were cultured on cassava flour, starch, wheat flour based media in order to explore the possibility of using them as substitutes to agar as a solidifying agent normallyused in tissue culture media preparation. The cassava flour, starch and wheat flour were used inplace of agar at a rate of 120, 160, 200 and 5240g/l of media which was prepared using stock solutions of Murashige and skoog. Use of paper bridges with liquid media was also investigated and agar based was prepared as an overall control. The findings indicate that agar can be effectively substituted by starch (120-200 g/l of media) and paper bridges in initiation cultures for cassava to give equally good results, while reducing on the costs. These findings are very promising for developing countries where procurement of tissue culture consumables can be very expensive. |
