The biochemical mechanism of the vrn1 gene action in the vernalization response in two isogenic lines of wheat, spring (D) and winter (C) was demonstrated to involve a higher rate of unsaturated phospholopid synthesis during 5-6 week period of growth at 2 °C in the winter genotype (C) compared to the spring (D). To further characterize the phospholipid metabolism in the two genotypes, incorporation of [32P]-orthophosphate and [14C]-acetate into tissue lipids of the two genotypes was followed during germination and growth at 2 °C and 25 °C. The pattern of [32P]-orthophosphate and [14C]-acetate into lipids of Triple Dirk (D) and (C) grown at 2 °C showed clearly that there was no significant difference in phospholipid synthesis up to 5 weeks of growth. However during the critical vernalization period of 5-6 weeks, the winter genotype increased linearly its rate of [32P] incorporation into phospholipids while the spring genotype with no vernalization response showed nearly a four-fold decrease in its rate of [32P] incorporation. During the same period, the 14C incorporation increased four-fold in the winter genotype with greater proportion of the unsaturated fatty acids. These changes confirm and extend the thesis that the biochemical mechanism of vrn1 gene function appears to act by stimulating the synthesis of unsaturated membrane phospholipids during the critical 5-6 weeks period of vernalization. |
