An attempt was made to chronologically examine the first meiotic events of caprine oocytes matured in vitro. The oocytes were harvested from caprine ovaries of abattoir origin by puncturing visible follicles with 18G hypodermic needle. Cumulus oocyte complexes were incubated for 6,12, 18,24 and 30 hr at 38°C, 95% relative humidity and a gaseous mixture of 5% COz in air. TCM-199 supplemented with 20% heatinactivated oestrous goat serum was used as culture medium. Subsequently, oocytes were fixed, stained with a?to-orcein and analysed for their chromosome configuration. GV stage was observed in 55,55% oocytes at 0 hr. It gradually decreased to 37.38, 11.34, 3.77, 8.90 and 4.38% at 6, 12, 18, 24 and 30 hr of culture. GVBD, an intermediate stage between GV and metaphase l, was the dominant configuration at 6 hr of incubation. Anaphase 1 and telophase 1 stage, first observed at 6 and 12 hr of incubation, respectively, were the rare stages of short duration in all the groups. Meiotically mature oocytes (metaphase II) first observed at 12 hr reached the highest level (54.79%) at 24 hr incubation. From the chronology of the first meiotic events, it is clear that maturation of caprine oocytes is complete within 24 hr of culture in vitro. |
