Wild species of Manihot have been cultivated in vitro for conservation proposes as well as for germplasm exchange and distribution among institutions. However, there is not a single culture medium to keep the large number of species of the genus under in vitro conditions because of particular requirements of each species. In the present report, a simple culture medium for wild species and rooting induction treatment was used to introduce new species in vitro, as well as rooting of previously in vitro cultivated plants without root system. The protocol for in vitro culture includes the use of activated charcoal in the medium and the treatment of explants with 2.5mM of IBA. Using this treatment, we were able to introduce 7 out of 8 Mexican wild species into culture and transfer 4 species cultivated in vitro with no roots to the greenhouse. |
