Forty-eight mature Cheviot ewes were allocated randomly in equal numbers to a control diet based on equal parts of goats and sugar beet shreds or the same diet with 3% or 6% herring oil. The diets were given from 22 d before egg collection, which occurred 5 d after intrauterine AI. All ewes received a 12 d progestogen sponge (0.3 g Cronolone; Intervet, UK) and 9 mg oFSH (Ovagen, ICP, UK) in 8 equal injections every 12 h from 60 h before to 24 h after sponge removal. Ovulations and large follicles (>8 mm) were recorded at egg recovery via mid-line laparotomy. Embryos were cultured in SOF with 0.4% w/v BSA (5% CO2, 5% O2; 38.5°C) for up to 48 h; development in vitro was recorded every 24 h. After 48 h culture, cell nwnbers (nuclei counts; Hoechst 33342 stain) and protein synthesis (3H-phenylalanine incorporation into protein; cpm) in blastocysts were recorded. Estradiol concentrations were determined in plasma collected at Al. Dietary effects on ovulations and embryo yields (square-root transformed), blastocyst development and grade (arcsine-transformed), cell nwnbers and protein synthesis (log- transformed) and zona-intact diameters were tested by ANOVA. Ovulations and embryo yields were unaffected by dietary lipid but estradiol concentrations increased (P<0.05), and were associated with a tendency (P=0.08) towards more large follicles at egg collection. Supplementary fish oil tended to suppress the cate of embryo developrnent and reduced blastocyst diameter (P=0.07) and cell nwnber (P<0.05) but not protein synthesis.
