One of the most important curculionids causing yield losses in soybean fields belongs to the genus Sternechus. The species of this genus are distributed in the Neotropical region. In the last 14 years, several authors have cited the presence of S. pinguis in Argentina, while Brazilian papers cite S. subsignatus. At present, no record of S. pinguis have been found in Brazil. The purpose of this paper is to clarify the identify of the Argentinean and Brazilian specimens and determine molecular polymorphism at mtDNA level. Samples os Sternechus were taken to verify the identify trough the analysis of morphological characters and comparisons of fragments from the mtDNA. Samples of the putative S. subsignatus and S. pinguis were taken in December of 2003 and January and February of 2004, from Monte Redondo, (Tucumán, Argentina), Mauá da Serra (PR), Warta (PR), São Desiderio (BA) and Samambaiba (MA). The mt DNA was amplified by polymerase chain reactions using the following primers LR-J-12887-LR-N-13398, LCO1490-J1514-HCO2198-N-2175 and CYT BF-CYT- BR. PCR products were treated with restriction enzymes Rsa I, Hae III, Pst I, Bam HI, Sma I, Apa I, Hind III and Taq I. The PCR was performed using a PTC 200 thermal cycler (MJ Research) programmed for 34 cycles of 94oC for 1 min, 48 or 55oC 1 min (depending on the primer) and 72oC for 1:30 min with a final extension at 72oC for 5 min. The studies based on morphological characters suggest that specimens from Argentina belong to the same species found in Brazil. The lack of molecular polymorphism between the specimens in other evidence that the populations belong to the same species.
