Abstract: Particle bombardment offers a tissue- and genotype-independente transformation technique that can be used to circumvent host strain restrictions sometimes encountered when using Agrobacterium-based transformation methods. We have previously developed an Agrobacterium-based transformation system for cassava. Now we present a novel protocol for Particle Inflow Gun (PIG) - mediated transformation, developed for use in combination with the organogenesis-based shoot regeneration system from somatic cotyledons. The parameters for transformation conditions have been partially optimised using transient GUS assays. The in vitro performance of different cassava cultivars after particle bombardment has been evaluated, and the frequencies of plant regeneration after bombardment and selection in cassava have been significantly improved by optimising the protocols further. The parameters partially optimised so far include the amount of DNA used, the flying distance of the microprojectiles, the duration of pre-and postplasmolysis treatments and the composition of the regeneration medium used after bombardment. |
