Abstract: In genetic engineering of cassava, tissue specific promoters are of prime importance. Therefore a cassava genomic library was constructed in the cosmid pBIC20 and will be screened with storage root specific probes in order to isolate cassava root specific genes and the corresponding promoters. A first screen of this library with a partial cDNA sequence of the cassava branching enzyme (BE), which is suppoded to be expressed specifically in the storage root, resulted in the isolation of a gene that partially showed 70% homology to the cassava branching enzyme. Further sequencig and the comparison to BEs from other plant species may confirm the possible existence of a second BE isoform in cassava. Expression studies with Northern blots will show if the isolated genes is well driven by a storage root specific promoter. A storage root cDNA library containing 1.5 10 pfu and cDNAs with sizes from 600 to 400 bp was constructed. In order to isolate more storage root specific cDNAs a differential +/- screen was perfomed. Identical replica filters, contaning about 30 000 pfu, were either probed with ³²P-dATP labelled storage root mRNA or with ³²P-dATP labelled leaf mRNA. By comparison of the replica filters more than 80 cDNAs could be identified which might have storage root specifi expression. It was possible to assign these cDNAs to at east 4 different groups. They will be sequenced and will then be assessed for the specificity of their expression. |
