Abstract: The objective of this study was to exanime mRNA transcription and enzyme activity of phenylalanine ammonia lyase (PAL) and peroxidase in cassava during root post-harvest deterioration. Cassava roots were transversely cut into 3.5-4.0 cm thick slices, and incubated in the dark on moistened paper towels in covered plastic containers. From the cut root slices, two sub-sections were collected for analysis, the outermost laver of 0.5 cm, and an inner layer 0.5 cm thick, located 1 cm in from the outermost layer. The inner layers showed a gradual increase in PAL activity. In contrast, the increase in PAL activity in the outer layers was more rapid, reaching a maximum in the initial hours after injury. Transcripts were detected as early as 6 h post injury, and increased rapidly to a maximum at 12 h; however, by 24 h PAL transcripts were scarcely detectable and increased gradually thereafter up to the experiment. Total peroxidase activity started to increase 24 hors after wounding in both layers. However, the inner layers showed an increase in both soluble and ionically bound-peroxidases, whereas the outer layers showed a higher level of the ionically-bound form in comparison to the soluble form. When AIP, an inhibitor of PAL, was applied to the roots, there were no visible differences in the pattern of root deterioration. However, there were a decrease in both PAL transcription and enzyme activity. Furthermore, differences in the balance of peroxidases was also observed in roots treated with AIP.
